Metagenome from plant compost

LCC is a cutinase found in metagenome from plant compost. It can degrade PET into terephthalic acid and ethylene glycol as the major products. The enzymatic activity of LCC was found to attain maximum at pH 8.0 and 50 degrees celsius. At this condition, the rate of PET degradation is 1.45 mg/ 24 hours. Roughly 70% of maximal activity can also be achieved at 30 and 70 degrees celsius. The specific PET-degrading activity of LCC at pH 8.0 and 50 degrees celsius was determined to be 12 mg/ hour/ mg of enzyme. LCC has a half-life of 5 hours, 80 minutes, 40 minutes and 7 minutes at 50, 60, 70 and 80 degrees respectively. It does not require divalent metal ions for activity. However, LCC is inhibited by E600 and LCC with signal peptide may not work properly in E. coli cells.
